I just primered the subframe to my 400ex with rust oleum wet sandable primer. Its dry now. Do I just lightly scuff the surface with a wet peice of sandpaper and then paint it?

The term wetsanding is usually associated with using a really fine grit paper somwhere round 800-higher and keeping it wet while you sand. This helps keep scratching to a minimum at that points so that the paint lyies flat. You are correct though and you should follow this every step of you painting.

Primer > sand
paint > sand
Paint > sand

Clear

At least this is how I do it everyone may have their own process.
 
You can just wet the paper, or squeze water out of a sponge, I have even seen people let a hose run over the area they are sanding.
The more time you spend preping and sanding, the better it will turn out.
Body work is meant for people with patience :)
